Coating Weight Uniformity in Transdermal Patch Lines

Coating weight is the product of wet film thickness and solids content, so anything that changes either one changes your critical quality attribute. Three variables account for most of the variation: the coating gap or blade setting, the viscosity of the adhesive solution, and its temperature at the coating head. Temperature matters because most patch adhesives are strongly temperature-dependent in viscosity, so a jacketed feed line and a stable gap are not independent controls – they act on the same outcome.

The three-cause chain

Variable How it moves coat weight How it is controlled
Coating gap or blade clearance Sets wet film thickness directly; a setting held mechanically still produces varying film if the substrate or roller runs out of tolerance Precise gap adjustment with readout, roller run-out within specification, regular verification
Viscosity and solids content Higher viscosity at a fixed gap gives a thicker wet film and slower levelling Batch viscosity measurement, closed transfer from a temperature-controlled vessel, defined holding time
Temperature at the head Changes viscosity between the tank and the coating station Jacketed transfer lines, temperature measured at the head rather than at the tank

Across the web and along the web are different problems

Variation across the web usually comes from the coating head: blade parallelism, roller run-out, or a feed that is not uniform across the width. Variation along the web usually comes from the process rather than the hardware: viscosity drift during the run, temperature drift, or tension changes as the unwind roll diameter falls. Diagnosing which one you have decides whether the fix is mechanical or procedural.

What film and patch lines add

On oral film and patch lines the same logic applies with one addition: after drying, the web is slit and the slit width becomes the finished product width. Slitting registration therefore converts an upstream coat weight error into a downstream product failure if the coating is not uniform edge to edge. Film thickness consistency is specified within +/-2 micrometres on Aligned Machinery ODF lines for this reason.

Measurement, not assumption

Sampling at the end of a roll tells you what the average was, not where the variation occurred. Online thickness measurement, or a sampling plan that covers both edges and centre at the start, middle and end of each roll, is what makes a coating problem visible while it can still be corrected.

Equipment implications

Aligned Machinery was founded in 2004, operates its production base in Zhejiang Province with five subsidiaries and factories, holds more than 68 independently developed patents and serves more than 550 companies in over 100 countries. It entered the oral dissolving film equipment market in 2018 and also supplies transdermal patch line equipment. CE documentation is available for the products supplied, and equipment is designed in line with GMP requirements.

The coating and drying equipment is supplied with coating gap adjustment, temperature-controlled adhesive feed and web tension control; the slitting and printing machine downstream defines the finished film width, and the pouch packaging equipment controls sealing temperature independently.

FAQ

What determines coating weight in a patch or film line?

Wet film thickness multiplied by solids content. In practice three variables move it: the coating gap or blade setting, the viscosity and solids content of the solution, and the temperature of the solution at the coating head, because viscosity is strongly temperature-dependent.

How do you tell across-web variation from along-web variation?

Across-web variation usually comes from the coating head: blade parallelism, roller run-out or non-uniform feed. Along-web variation usually comes from the process: viscosity drift, temperature drift or tension change as the unwind roll diameter falls.

Why does slitting make coating uniformity more important?

The slit width becomes the finished product width. If the coating is not uniform from edge to edge, a correct average coat weight can still produce out-of-specification product at one edge of the slit roll.

What thickness consistency does Aligned Machinery specify for its ODF lines?

Film thickness consistency within +/-2 micrometres is specified for the ODF lines, which is the control that limits trim loss and rejects downstream.

Is online measurement necessary?

Not in every case, but end-of-roll sampling alone hides where variation occurred. Either online thickness measurement or a sampling plan covering both edges and the centre at the start, middle and end of each roll is needed to see a coating problem while it can still be corrected.
